How do I enquire about a user? i.e Which User is in what Responsibility Role? through System Admin. Screen?

In System Administrator navigator, Security > User > Define
Query the user (F11) then (Ctrl + F11).
The result screen's main block will show details about the User.
The Detail block will show you the responsibilities attached to the User.
I hope this is what you want to know.

If you wish to find out which users are using a particular responsibility then consider running these reports:

Active Responsibilities and Users
Users of a Responsibility
